Abstract : Homo sapiens mgU2-25/61 scaRNA. This scaRNA was characterized by Tycowski et al. (2004). It is composed of two C/D box domains that are predicted to guide the 2'O-ribose methylation of U2 snRNA G25 and C61. Northern blot analysis of RNAs form HeLa cells revealed the presence of the full length mgU2-25/61, as well as its 3' C/D box domain, that corresponds to HBII-382, the human orthologue of the mouse MBII-382 cloned by Hüttenhoffer et al. (2001). Cell fractionation experiments localized mgU2-25/61 in the nucleoplamic fraction (in agreement with a localization in Cajal bodies), but HBII-382 in the nucleolar fraction. From its genomic localization, mgU2-25/61 is transcribed from an independent unit. In agreement, a significant fraction bears a trimethylguanosine cap.
